减压内部沸腾法提取壶瓶枣多糖及其脱色工艺的研究

Extraction of Polysaccharides from Ziziphus Jujube Mill. cv. Hupingzao by Reduced Pressure Boiling Method and Its Decoloration Technology

摘要: 对减压内部沸腾法提取壶瓶枣多糖及其脱色工艺进行了研究。以蛋白质和多糖得率为指标,通过正交试验优化得到的最佳工艺条件为:体系内温度60 ℃,液料比为20:1(mL:g),时间为30 min,此时真空度为80 kPa,外界温度为70 ℃,蛋白质和多糖得率分别为0.13%和2.60%。与传统热浸提相比,其得率分别提高了18.18%和23.22%。对10种脱色材料的筛选结果表明D900型大孔吸附树脂是理想脱色材料,通过正交试验优化得到的D900型大孔吸附树脂脱色优化工艺参数为D900添加量3.5%,时间20 h,pH值11.7,温度40 ℃,此时脱色率、脱蛋白率和多糖保留率分别为81.94%、38.19%和87.42%。

Abstract: Extraction of polysaccharides from Zizyphus jujube Mill. cv. Hupingzao by reduced pressure boiling method and its decoloration technology were studied in this paper.According to protein and polysaccharide yield, the optimum technological conditions were studied by orthogonal test. The results showed that the optimum technological conditions were system temperature 60 ℃, water/solid ratio 20:1(mL:g), time 30 min, vacuum degree 80kPa, and external temperature 70 ℃. Under these conditions, the protein and polysaccharides yields were 0.13% and 2.60%, respectively. Compared with traditional water extraction, the protein and polysaccharides yields were improved by 18.18% and 23.22%, respectively. According to the decolorization ratio, deproteinization ratio and retention rate of polysaccharides, macroporous adsorption resin D900 was the suitable one among 10 kinds of decolorization materials. Orthogonal test was employed to optimize the decolorization conditions. The results indicated that optimum decolorization conditions were additive amount of D900 3.5%, time 20 h, pH value 11.7 and temperature 40 ℃, respectively. Under these conditions, the decolorization ratio, deproteinization ratio, and retention rate of polysaccharides were 81.94%, 38.19%, and 87.42%, respectively.
