Abstract: Homogenate extraction conditions of polysaccharides from Gardenia jasminoides Ellis were studied. Single factors including extraction time, alcohol concentration, and liquid to solid ratio were investigated separately. The response surface analysis method followed by verification test was conducted for obtaining the appropriate operation conditions. Results showed that the optimal conditions of polysaccharides extraction were as follows: extraction time 42 s, alcohol concentration 5% and liquid to solid ratio 27:1(mL:g). Under the above conditions, the yield of polysaccharides was 6.38%. It matched with the predicted yield. Yield of polysaccharides using homogenate extraction was 32.5% higher than that in traditional heat refluxing extraction. Hence, homogenate extraction is a good alternative to extract polysaccharides from G.jasminoides.
