分离制备高纯度中性木低聚糖的研究

摘要: 研究了木聚糖酶定向酶解木聚糖水解液中主要成分在聚丙烯酰胺凝胶柱层析时被洗脱分离的顺序,确定了较佳的洗脱速度和进样量。结果表明,酶蛋白、木质素是木低聚糖酶解液中分子量最大的物质,在层析时最先被洗脱出来。酶解液中含有 12.8%的酸性低聚糖,在洗脱时和木八以上的可溶性聚糖混合在一起。用 55cm×7.5cm层析柱在 50℃、以脱离子水为流动相的分离条件下,快速制备高纯度中性木低聚糖的适宜洗脱速度和进样量分别为 800mL/h、120mL。

Abstract: The eluting orders of the main constituents in enzymatic hydrolysate of xylan on polyacrylamide column were studied,and the suitable elution flow rate and injection volume were determined.The results showed that protein and lignin were first eluted because of their highest molecular weight in hydrolysate.There were 12.8% acidic oligosaccharides in hydrolysate,eluted with the soluble xylan of DP>8 together.Using the proper elution flow rate and injection volume at 800mL/h and 120mL,respectively,high purity neutral xylo-oligosaccharides were rapidly isolated using 55cm×7.5cm polyacrylamide gel column,distilled water as flow phase,at 50℃.
