耐热型木聚糖酶制备工艺的研究

摘要: 以木糖渣为碳源,利用耐热子囊菌(Thermoascus aurantiacus)液体发酵生产木聚糖酶,确立了最佳工艺参数为:木糖渣质量浓度4%、培养温度45℃、转速180r/min、培养基初始pH值4.8、初始C/N为7.1,木聚糖酶活力可达900U/mL。酶学性质研究表明,该酶反应的最适温度为75℃,最适pH值为4.8。该酶的耐热性强,在75℃下保温lh,酶活力剩余85%以上。

Abstract: Preparation of xylanase by Thermoascus aurantiacus on corn cob residue was studied.Optimal fermentation parameters were:corn cob residue 4%;rotating speed 180 r/min;initial pH value of medium 4.8%;culture temperature 45℃;C/N of medium 7.1.Xylanase activity in the broth reached 900 U/mL.Optimal temperature and pH value for the xylanase reaction were 75℃ and 4.8,respectively.The enzyme preparation was thermo stable and had a good application potential.
