均匀设计法优化桦木醇的超临界二氧化碳萃取工艺

Optimization of the Extraction of Betulin by Supercritical CO2 Using Uniform Design

摘要: 利用均匀设计法优化了超临界CO2萃取(SCDE)桦树皮中桦木醇的工艺.采用均匀设计法考察了萃取温度、萃取压力、萃取时间对桦木醇收率的影响,并用高效液相色谱法测定了萃取物中桦木醇的含量.试验结果表明,在萃取温度32℃、萃取压力8MPa、萃取时间1h、CO2流量为20kg/h的条件下,SCDE的桦木醇得率为16.96%,萃取物中桦木醇含量达到74.88%.与其他提取方法相比,SCDE法尽管得率略低,但选择性好且无环境污染,具有一定的应用前景.

Abstract: The technique for the extraction of betulin by supercritical CO2 (SCDE) was optimized using uniform design. The effects of extraction temperature, pressure and time on betulin yield were discussed, and betulin content was determined by HPLC. A yield of 16.96% and purity of 74.88% were achieved under the optimal conditions as follows: extraction temperature 32℃, extraction pressure 8 MPa, extraction time 1 h and CO2 flow rate 20 kg/h. Compared with other extracting methods, though the yield of SCDE was lower, the selectivity was higher together with no environmental pollution, which may achieve an applicable prospects for SCDE of betulin.
