流动注射化学发光法测定茜草总黄酮清除羟自由基能力的研究

Determining the Ability of Madder Flavonoids Scavenging Hydroxy Radical Using Flow Injection Chemiluminescence Method

摘要: 以茜草叶醇提取的茜草总黄酮为原料,采用流动注射化学发光法,以Vc为对照,考察不同浓度、时间、光照及加热处理下茜草总黄酮清除·OH 的能力。在0.000 5~0.1 g/L 质量浓度范围内,茜草总黄酮对·OH 的清除能力与其浓度呈正相关,在质量浓度为 0.1 g/L 时,对·OH 清除率高达 97.23 %;放置 58 h 后,茜草总黄酮对·OH 的清除率(94.18 %)比Vc(17.6 %)高了76.58个百分点;132 W 灯照 58 h 后,茜草总黄酮对·OH 的清除率下降了2.79个百分点,Vc对·OH 的清除率下降了88.55个百分点;在加热煮沸处理 10 h 后,茜草总黄酮的耐热性较强,10 h处理后,其对·OH 的清除率(88.4 %)比Vc(65.07 %)高了23.33个百分点。因此,茜草总黄酮的天然抗氧化能力较强,具有一定的开发应用价值。流动注射化学发光法灵敏度高、测定快速、操作简单和重现性好,可应用于茜草总黄酮清除超氧阴离子的能力评价及精制茜草总黄酮的抗氧化效果研究。

Abstract: The influences of concentration, time, light and heating processing on scavenging ·OH were investigated by flow injection chemiluminescene method comparing with Vc. In the concentration of 0.000 5~0.1 g/L, the scavenging ability to ·OH was a positive corelation with the concentration of madder flavonoids,when the concentration was 0.1 g/L,the clearance rate of ·OH was 97.23 %; After 58 h of storge, the scavenging rate of ·OH by madder flavonoids was higher than the Vc by 76.58 % percentage points; After irradiated by a 132 W lamp for 58 h, the scavenging rate of ·OH by madder flavonoids decreased by 2.79 percentage points, Vc decreased by 88.55 percentage points; After the treatment in boiling water for 10 h, madder flavonoids have strong heat resistance, the scavenging rate of ·OH was higher than the Vc by 23.33 percentage points.Therefore,madder flavonoids have good ability of natural antioxidant,and possess a certain value of development and applications. Flow injection chemiluminescence method has the advantages of high sensitivity, rapid determination, simple operation and good reproducibility, and it can be used to evaluate the capacity assessment of superoxide anion and antioxidant effect of madder flavonoids.
