黑荆树皮单宁不同级分对水解酶的抑制作用

摘要: 通过对黑荆树皮单宁进行醇沉纯化和溶剂萃取分级,可得到分子量分布范围不同的单宁级分。各级分对 5种水解酶均表现出不同程度的抑制作用,具有一定的选择性。其抑制能力与抑制剂常数Ki值有关,分子量大的单宁级分,Ki值小,对酶活性的抑制作用大,同时,证明了黑荆树皮单宁对纤维素酶的抑制作用属于非竞争性抑制。

Abstract: Tannin fractions with different molecular weight distributions were obtained by purification of alcohol preci pitation and fractionation of solvent extractions of black wattle bark tannins. The data observed in the experiments indicated that tannin fractions possess varying inhibitory effects on activity of hydrolases, showing selective inhibition. Based on comprehensive investigations of cellulase, it was displayed that the inhibitions are related to the inhibitor constant Ki. When the molecular weight of the fractions is higher, Ki value is lower and the inhibitory effect is stronger. Meanwhile, the inhibition of the fractions on the cellulase was proved to be a noncompetitive one.
