Studies on Extraction and Purification of Flavonoids from Phyllostachys heteroclata Oliver

Abstract: Flavonoids from Phyllostachys heteroclata Oliver were extracted by methods of waterbath and ultrasonic extractions.The optimum extraction conditions were obtained by single factor and orthogonal tests.Different kinds of macro-reticular resins were tested for purification.The results showed that the yield of flavonoids was 1.65% by waterbath extraction under the conditions of 60% ethanol, 50℃,1:20 solid-liquid ratio and 1.5 h.Order of the factors which affected waterbath extraction was: ratio of soild to liquid>concentration of ethanol solution>extraction temperature>extraction time. The yield of flavonoids was 1.27% by ultrasonic extraction under the conditions of 60% ethanol, 1:15 solid-liquid ratio and 30 min. Order of the factors which affected ultrasonic-assisted extraction was: extraction time>volume fraction of ethanol solution>ratio of soild to liquid.It was found that AB-8 macro-reticular resin was superior for the purification of flavonoids,and 70% ethanol was a good eluant.
